KRAS is mutated in roughly a quarter of all human cancers, and G12D is the most common variant of all — yet it has been among the hardest to drug.

BX-101 is an oral targeted degrader designed to remove mutant KRAS G12D protein rather than block it, with selectivity over wild-type KRAS. Removing the protein should also avoid the rebound signalling that limits inhibitors.

Around a third of people with Crohn's disease develop fibrostenotic complications, and no current therapy is designed for them.

BX-204 is an oral TL1A antagonist. TL1A sits upstream of both the inflammatory and the fibrotic arms of intestinal disease, which is why we selected it over the better-trodden cytokine targets — and why our trials stratify patients by intestinal TL1A expression.

ALS has a median survival of two to five years from diagnosis, and disease-modifying options remain very limited.

BX-308 is an intrathecally delivered antisense oligonucleotide against a genetically validated ALS target. Neurofilament light is built into the design from first-in-human onward, so we will know early whether we are affecting the disease process.

BX-412 is a liver-directed siRNA that silences LPA transcription. Human genetics gives an unusually clean read on both direction and magnitude of effect here, which is exactly the kind of target a small company should be working on.
